چکیده

The expression of human epidermal growth factor receptor 2 (HER2) on the surface of individual exosomes secreted from human breast cancer cells was evaluated by on-chip microcapillary immunoelectrophoresis. A positive shift of the zeta potential was observed for exosomes secreted from HER2positive SK-BR-3 cells, whereas no difference was observed for exosomes secreted from HER2-negative MDA-MB-231 cells. These findings suggest that on-chip immunoelectrophoresis is a promising methodology for analyzing the surface HER2 protein on individual exosomes for future applications to lessinvasive HER2 testing.

On-Chip Immunoelectrophoresis of Extracellular Vesicles Released from Human Breast Cancer Cells

Extracellular vesicles (EVs) including exosomes and microvesicles have attracted considerable attention in the fields of cell biology and medicine. For a better understanding of EVs and further exploration of their applications, the development of analytical methods for biological nanovesicles has been required. In particular, considering the heterogeneity of EVs, methods capable of measuring i...

Comparing the Effect of Silybin and Silybin Advanced™ on Viability and HER2 Expression on the Human Breast Cancer SKBR3 Cell Line by no Serum Starvation

The polyphenol silybin has anti-oxidant and anti-cancer properties. The poor bioavailability of some polyphenols (flavonoids, and terpenoids) can be improved by binding them to phosphatidylcholine (phytosome technology). Many studies have focused on the most common phytosome, silybin-phosphatidylcholine, particularly for its hepatoprotective effects. However, in recent years, studies have also ...

Clinical application of a microfluidic chip for immunocapture and quantification of circulating exosomes to assist breast cancer diagnosis and molecular classification

Increasing attention has been attracted by exosomes in blood-based diagnosis because cancer cells release more exosomes in serum than normal cells and these exosomes overexpress a certain number of cancer-related biomarkers. However, capture and biomarker analysis of exosomes for clinical application are technically challenging. In this study, we developed a microfluidic chip for immunocapture ...

The effect of exosomes derived from human ovarian epithelial cancer cells on the secretion of AMH and Inhibin in granulosa cells

Exosomes are secreted by different types of cells and known as biological packages. Exosomes have significant role in intercellular communications and involved in the development and progression of various diseases such as cancer. Inhibin B and anti-mullerian hormone (AMH) are markers of granulosa cell tumors (GCT) and due to the role of exosomes in the progression of cancer, in this experiment...
